刨花润楠组织培养技术的初步研究

摘 要:

以刨花润楠种子为外植体, 采用丛芽发生途径, 初步建立其组培快繁技术体系。结果表明, GA3浸泡种子可显著提高发芽率, 缩短发芽时间。子叶节最易诱导不定芽, 诱导芽数最多。刨花润楠丛芽再生能力较弱, 培养过程中愈伤组织多, 整体情况较差。较为适宜的增殖方式为外植体在MS+6-BA 3.0 mg•L-1+IBA 0.01 mg•L-1+AgNO3 1.0 mg•L-1上预培养10 d, 再转入MS+6-BA 3.0 mg•L-1+IBA 0.01 mg•L-1中培养, 增殖系数可达3.52。生根培养基为1/2MS+IBA 0.2 mg•L-1, 生根率可达91%。

Primary study of tissue culture of Machilus pauhoi

Abstract:

Tissue culture of Machilus pauhoi were studied through clustered bud cultivation from seeds as explants. The results showed that the germination rate of Machilus pauhoi seeds was effectively improved by soaking the seeds with gibberellin. The cotyledon node was the best part for induction. The best way to proliferation was the plantlets pre-culture on the medium with AgNO3 (MS+6-BA 3.0 mg•L-1+IBA 0.01 mg•L-1+AgNO3 1.0 mg•L-1) for ten days, then transplanted into the same medium without AgNO3 (MS+6-BA 3.0 mg•L-1+IBA 0.01 mg•L-1), the proliferation coefficient was up to 3.52. The better rooting medium was 1/2MS+IBA 0.2 mg•L-1, the rooting rate was above 91%.
